Tiivistelmä
Tämän opinnäytetyön tavoitteena oli selvittää valon intensiteetin ja spektrin vaikutusta lehtimangoldin miniversojen kasvuun ja fenolisten yhdisteiden määrään. Työn tilaajana oli HAMK Bio-tutkimusyksikkö. Opinnäytetyön kirjallisuusosassa perehdyttiin julkaisuihin, jotka koskivat kasvien sekundaarimetaboliaa, fenolisia yhdisteitä ja valon vaikutusta niihin.
Opinnäytetyön tutkimuksellisessa osuudessa tehtiin kasvatuskoe, jossa lehtimangoldin (Beta vulgaris ssp. vulgaris var. cicla) versoja kasvatettin 17 vuorokauden ajan kolmessa eri valo-olosuhteessa. Matalan (~170 µmol/m²/s) ja korkean (~300 µmol/m²/s) valointensiteetin käsittelyissä valon lähteenä toimivat loisteputket. LED-valokäsittelyssä (LED300) yhdistettiin kaksi punaista (Hyper Red; 660 nm) ja kaksi sinistä (Deep Blue; 450 nm) LED-putkea ja valon intensiteetti säädettiin tasolle ~300 µmol/s/m². Valokäsittelyiden vaikutusta lehtimangoldin miniversojen kasvuun selvitettiin määrittämällä kasvien tuore- ja kuivapainot. Kasvinäytteet kuivattiin ja fenoliset yhdisteet uutettiin kiihdytetyllä liuotinuutolla. Kokonaisfenolipitoisuus määritettiin Folin-Ciocalteu -menetelmällä UV-Vis spektrofotometrillä.
Molempien korkeassa valon intensiteetissä (LP300 ja LED300) kasvaneiden miniversojen tuore- ja kuivapainot olivat suurempia kuin matalassa valon intensiteetissä (LP170) kasvaneiden miniversojen painot (p<0,05). Fenolisten yhdisteiden määrä oli korkein LED-valotuksessa (LED300) kasvaneissa versoissa ja ero oli tilastollisesti merkitsevä suhteessa matalan intensiteetin (LP170) versoihin (p<0,05). Keskinäisessä vertailussa korkean valon intensiteetin käsittelyiden (LP300 ja LED300) kokonaisfenolipitoisuuksissa ei ollut tilastollisesti merkitsevää eroa (p>0,05).
Tässä opinnäytetyössä havaittiin, että korkeampi valon intensiteetti lisäsi kasvua sekä loisteputki- että LED-valotuksessa. Lehtimangoldin miniversojen sisältämien fenolisten yhdisteiden määrään voidaan tämän opinnäytetyön tulosten perusteella mahdollisesti vaikuttaa kasvattamalla miniversoja spektrissä, joka sisältää runsaasti sinistä ja punaista aallonpituutta, kun valon intensiteetti on tasolla ~300 µmol/s/m². Valon intensiteetin ja spektrin vaikutusta lehtimangoldin kasvuun ja kokonaisfenolipitoisuuteen tulee tutkia vielä lisää, jotta löydetään valo-olosuhteet, joilla voidaan optimoida sekä sadontuotto että fenolisten yhdisteiden määrä. The aim of this practise-based thesis work was to investigate the effect of light intensity and spectra on the growth and the total phenolic content (TPC) of Swiss chard (Beta vulgaris ssp. vulgaris var. cicla) microgreens. The project was commissioned by the HAMK Bio Research Unit. The literature part of the thesis focuses on publications on secondary metabolism of plants, phenolic compounds and the effect of light on them. The practical part of the work was carried out by growing microgreens in different lighting conditions.
Swiss chard microgreens were grown for 17 days under three different light conditions. In low (~170 μmol/m²/s) and high (~300 μmol/m²/s) light intensity treatments, fluorescent lamps acted as the light source. In the LED light treatment (LED300), two red (Hyper Red; 660 nm) and two blue (Deep Blue; 450 nm) LED tubes were combined, and the light intensity was adjusted to ~300 μmol/s/m². The effect of light treatments on the growth of Swiss chard microgreens was investigated by determining the fresh and dry weights of the plants. The plant samples were dried, and the phenolic compounds were extracted by accelerated solvent extraction (ASE). The total phenolic content was determined by the Folin-Ciocalteu method using a UV-Vis spectrophotometer.
The fresh and dry weights of both microgreens grown at high light intensity (LP300 and LED300) were higher than those of microgreens growing at low light intensity (LP170) (p<0.05). The total phenolic content was highest in microgreens grown under LED exposure (LED300) and the difference was statistically significant compared to low intensity (LP170) shoots (p<0.05). In comparison, there was no statistically significant difference in total phenolic content between high light intensity treatments (LP300 and LED300) (p>0.05). Based on the results of this growth experiment, it was found that higher light intensity increased growth in both fluorescent and LED exposure. The total phenolic content in Swiss chard microgreens can possibly be increased by exposing them to blue and red wavelengths at the light intensity of ~300 μmol/m²/s. However, the effect of light intensity and spectra on Swiss chard growth and total phenolic content needs to be further studied to find light conditions that can optimize both growth and total phenolic content.
