Tiivistelmä
Panossyöttökasvatus on bioprosessiteollisuudessa käytetyin kasvatustyyppi. Se alkaa kuten panoskasvatus, mutta alkuperäisten raaka-aineiden loputtua kasvatukseen syötetään lisää ravinteita. Näin kasvatus pidentyy, minkä ansiosta solumassa moninkertaistuu. Korkeampi solumassa johtaa samalla myös korkeampaan tuottoproteiinin loppukonsentraatioon. Tuottoproteiinin korkea tuottotaso edellyttää kuitenkin usean parametrin optimointia, kuten syötettävien lisäravinteiden pitoisuutta ja syöttönopeutta, happamuutta, lämpötilaa sekä induktiohetkeä. Näiden parametrien optimointia hankaloittaa kuitenkin se, että tuotettavia proteiineja voidaan harvoin havaita reaaliaikaisesti. Tämän lisäksi tuottoproteiinin pitoisuuden mitttaamiseen tehtävät spesifiset määritykset voidaan joissain tapauksissa tehdä vasta jälkeenpäin puhdistusvaiheiden jälkeen.
Diplomityön tavoitteena oli kehittää menetelmä ja laitteisto tuottoproteiinigeenin ekspressiota ohjaavan promoottorin aktivoitumisen reaaliaikaiseen seurantaan Escherichia coli-panossyöttökasvatuksessa. Tämä tehtiin kehittämällä tuottoplasmidi, jossa kohdeproteiinin geenin lisäksi plasmidiin lisättiin vihreää fluoresoivaa proteiinia (engl. Green fluorescent protein, GFP) koodaava geeni. Kummankin proteiinin tuottoa ohjattiin oman lac-promoottori-operaattorinsa avulla, minkä johdosta indusoitaessa molemmat proteiinit tuottuvat samanaikaisesti. GFP:n mittaamiseksi suunniteltiin mittalaite, joka kykenee virittämään ja mittaamaan GFP:n fluoresoivaa valoa bioreaktorista reaaliajassa. Tämä toteutettiin kierrättämällä kasvatusta jatkuvatoimisesti mittalaitteen läpi käyttäen peristalttipumppuja. Valon mittaamiseen tutkittiin fotodiodin, kameran sekä pienikokoisen valomonistinputken soveltavuutta.
GFP ei vaikuta negatiivisesti kohdeproteiinin tuottoon, ja fluoresenssisignaali korreloi kohdeproteiinin tuottopitoisuuden kanssa, kun GFP:n maturaatioaika otetaan huomioon. Pienikokoinen valomonistinputki ja kamera olivat valoantureista herkimmät ja niiden avulla havaittiin lac-promoottori-operaattorin aktivoituminen kahdessa eri E. coli-panossyöttökasvatuksessa. Reaaliaikasta seurantaa ei saavutettu GFP:n kromoforin maturointiin vievästä ajasta sekä kierrätykseen kuluvasta viiveestä johtuen. Tulosten perusteella kehitettyä menetelmää voidaan kuitenkin käyttää proteeinituoton seurantaan ja optimointiin. Fed-batch fermentation is the most used process type in biopharmaceutical industry. It starts as a batch fermentation but when the original nutrients are depleted, new nutrients are fed to the bioreactor. This will prolong the process and high cell-density can be reached. Higher cell-densities will help producing higher concentrations of product. For attaining high concentrations of product numerous factors, such as feed strategy, pH, temperature, and induction time, need to be optimised. The optimisation is hindered by the lack of possibility for real-time monitoring of the target protein. Some specific assays for the target protein can only be done after purification steps.
The goal of the thesis was to develop a method and a device for real-time monitoring of lac-promoter activation in Escherichia coli fed-batch fermentation. A production plasmid was cloned containing the gene for the target protein and the gene for green fluorescent protein (GFP) with their individual lac-promoters. This dual-promoter structure guarantees the maximum control and concomitant gene expression. A measurement device capable of exciting and measuring the emitted fluorescence was constructed for real-time monitoring of GFP. The culture broth was continuously recycled through the measurement device using peristaltic pumps. Photodiode, camera and photomultiplier tube (PMT) were compared as detectors.
GFP does not affect negatively to the production of target protein. Fluorescence signal correlates with the product concentration when the maturation delay of GFP chromophore is taken into account. PMT and camera were the most sensitive detecting GFP in saline solution and were used in two separate fed-batch E. coli fermentations for monitoring the lac-promoter activation. Both detectors were successful in detecting the protein production. Real-time monitoring was hindered by the maturation time of GFP chromophore and recycling delay. Based on the results, the method can be used for monitoring protein production, albeit not in real-time, and for process optimisation.
