Tiivistelmä
Tämän opinnäytetyön tarkoituksena oli optimoida automatisoitu pudistusmenetelmä synteesin välituotteelle käyttämällä CombiFlash EZ Prep -laitetta. Työssä keskityttiin poistamaan tai korvaamaan manuaalisesta menetelmästä haitalliset liuottimet vähemmän haitallisilla liuottimilla. Automatisoidulle menetelmälle haluttiin löytää parhaat mahdolliset laitteistoparametrit, pylväsmateriaalit ja näytteen injektoimistavat jokaiselle välituotteelle.
Työssä tehtiin 40 flash-kromatografia-ajoa kahdelle eri välituotteelle, joista suurin osa oli yhdisteelle 9. Työn aikana ensimmäisen välituotteen puhdistusmenetelmä saatiin optimoitua niin pitkälle, että päästiin tekemään tuotantomittakaavan ajo. Toisen välituotteen kanssa saatiin tehtyä muutama pienempi ylöspäin skaalaus, ajot jäivät kuitenkin vähemmälle ajan puutteesta johtuen. Näiden yhteydessä kromatografia-ajoissa kokeiltiin kolonnin uudelleenkäyttöä, näytemäärää kolonnissa ja kolmea eri injektoimistapaa.
Ennen kromatografia-ajoja oikeanlaista eluenttia etsittiiin ohutkerroskromatografian avulla kokeilemalla erilaisia liuotinseoksia ja niiden suhteita. Ohutkerroskromatografialevyistä nähtiin lähimmät epäpuhtaudet tuotteen ylä- ja alapuolella, joita käytettiin apuna kromatografia-ajojen optimoinnissa. Kromatografia-ajojen jälkeen tuloksia analysoitiin kromatogrammin lisäksi myös ohutkerroskromatografian avulla.
Kromatografia-ajoihin löydettiin vähemmän haitallinen vaihtoehto, joka oli etyyliasetaatin ja heptaanin yhdistelmä. Ensimmäiselle välituotteelle paras erottelevuus saatiin lineaarisella gradientilla ja pidennetyllä ajo-ohjelmalla. Toiselle välituotteelle todella loiva lineaarinen ajo oli paras vaihtoehto vaikka menetelmän optimointi kyseiselle välituotteelle jäi kesken. Injektoimalla valmisnäytepatruunan avulla saatiin toistettavuutta tuloksiin ja erottelevuus oli hyvä. Pienemmillä määrillä tuotetta kolonnien uudellenkäyttö oli mahdollista ja erottelevuus oli hyvä kun tuotetta oli alle kolonnin suositellun maksimi näytemäärän.
Yhdisteen 9 tuotantomittakaavan ajossa tuotteen joukosta löytyi vähän epäpuhtauksia, mutta ajo oli silti toivotun saantoprosentin sisällä. Yhdisteen 9 tuotantomittakaavan ajo oli onnistunut, eikä sen automatisoitua menetelmää tarvinnut lähteä enää parantamaan. Yhdisteen 10 ajot olivat myös onnistuneet, vaikka epäpuhtauksia tulikin yhtä paljon kuin manuaalisella puhdistusmenetelmällä. Testauksia voisi tästä eteenpäin jatkaa vielä yhdisteellä 10 aina tuotantomittakaavaan asti. Kun yhdisteen 10 automatisointi olisi optimoitu voitaisiin siirtyä tutkimaan muiden synteesivälituotteiden automatisointia. The objective of this thesis was to optimize an automated purification method by using CombiFlash EZ Prep equipment. The thesis focused on removing or replacing harmful solvents from the manual method with less harmful solvents. For the automated method the aim was to find the best possible equipment parameters, column materials, and sample injection methods for each intermediate product.
The study involved 40 flash-chromatography runs for two different intermediate products, whereby most of them were for compound 9. During the study, the first intermediate product purification method testing progressed such that a production scale run was performed. With the other intermediate product testing, a few smaller scale-up runs were conducted due to lack of time. During these chromatography runs column reuse, amount of sample in column, and three different injection methods were also tested.
Before the automated chromatography runs, a less harmful solvent was found from the automated method using thin-layer chromatography by trying different solvent mixtures at different mixture ratios. Thin-layer chromatography plates showed the closest impurities above and below the desired product. These results were used later in the optimization of flash-chromatography runs. After the automated run was performed it was analyzed with a chromatogram and thin-layer chromatography.
The less harmful solvent was found to be a mixture of ethyl acetate and heptane. For the first intermediate product, the best resolution was obtained with a linear gradient and an extended run program. For the second intermediate product, a less steep linear run was the best option even though the optimization of the method for that intermediate product was not completed. An injection with a ready-to-use sample cartridge gave result reproducibility and a good resolution. With smaller amounts of product, it was possible to reuse the columns and the resolution was good when the amount of sample was below half of the recommended maximum for the column.
In the production scale run of compound 9, few impurities were found among the product and the run was still within the desired yield percentage. The production scale run of compound 9 was successful and its automated method no longer needed to be improved. The automated runs of compound 10 were also successful although the purity of the product was the same with manual methods. From this point, the testing could be continued with compound 10 all the way up to the production scale. Once the automation of compound 10 has been optimized, the automation of other intermediate synthetic products could be tested next.
