Abstract
In this thesis a novel microfluidic perfusion chip for ex vivo brain slice imaging was designed and fabricated and used to image neuronal activity in acute coronal mice brain slices. Microfluidic principles were used for more precise control of pharmacological agents and to ensure the viability of the slices. The perfusion chips were fabricated by replication moulding in a three-step process from thiol-ene. GCaMP6-viral vectors were injected into mice hippocampus in stereotaxic operations. After recovery time the GCaMP6 transduced and transgenic mice were sacrificed and ex vivo brain slices were prepared and stabilized. The ex vivo slices were placed in the perfusion chamber and stimulated with depolarizing potassium chloride step while being imaged with two-photon microscope. Recorded videos were analysed, and statistical testing was used to prove the effectiveness of the perfusion chamber and the methods of ex vivo brain slice preparation for studying the activation of neurons in acute brain slices.

Tässä lopputyössä suunniteltiin ja valmistettiin uudenlainen mikrofluidistinen perfuusiosiru, jota käytettiin koronaalisten akuuttien ex vivo aivoleikkeiden neuroniaktiivisuuden kuvantamiseen. Mikrofluidistisia periaatteita käytettiin tarkempaan lääkeaineiden kontrolliin ja varmistamaan aivoleikkeiden elinvoimaisuus. Perfuusiosirut valmistettiin tioleenistä kolmivaiheisella replikaatiomuottivalmistusmetodilla. GCaMP6 virusvektoreita injektoitiin hiirien hippokampukseen stereotaksisessa kirurgiassa. Palautumisajan jälkeen GcaMP6 transdusoituneet hiiret ja transgeeniset hiiret lopetettiin ja ex vivo aivoleikkeet valmistettiin ja stabiloitiin. Ex vivo leikkeet asetettiin perfuusiokammioon, jossa niitä stimuloitiin depolarisoivalla kaliumkloridi toimenpiteellä, samalla kun niitä kuvannettiin kaksi-fotoni mikroskoopilla. Kuvatut videot analysoitiin ja tilastotieteellisesti todistettiin perfuusiokammion ja ex vivo aivoleikemetodien toimivuus akuuttien aivoleikkeiden neuroniaktiivisuuden tutkimiseen.
