Cellen har välutvecklade system för att hantera en varierande omgivning och olika typer av stress. Bland dessa skyddssystem återfinns värmechockresponsen som regleras med hjälp av värmechockfaktorerna HSF1 och HSF2, som fungerar som transkriptionsfaktorer och aktiverar transkription av chaperoner som till exempel Hsp70. Värmechockresponsen aktiveras också vid oxidativ stress. Avhandlingens syfte var att undersöka inverkan av oxidativ stress på proteinnivåerna av HSF1 och HSF2 och deras bindningsmönster till DNA. Oxidativ stress skapades i K562-celler med H2O2 eller menadion och de relativa proteinnivåerna jämfördes med Western blot-analys. Cellerna behandlades också med proteasominhibitorn MG-132 för att undersöka proteasomens inverkan på proteinnivåerna av HSF1, HSF2 och Hsp70.
Oxidativ stress påverkade lösligheten av HSF1 och HSF2, eventuellt så att de bildar olösliga kromatinaggregat. Oxidativ stress inhiberade induceringen av Hsp70 vid proteasominhibering, både på mRNA- och proteinnivå via en okänd mekanism. HSF1 och HSF2 band promotorn för Hsp70 vid oxidativ stress och proteasominhibering, men varken mRNA eller Hsp70-protein producerades mer än normalt, vilket var överraskande. Oxidativ stress har en inhiberande inverkan på värmechockresponsen i samband med andra stressfaktorer, eventuellt till förmån för cellens andra stresshanteringssystem. The heat shock response, regulated by heat shock factors, is a highly conserved mechanism acting to maintain protein stability during cell stress. HSF1 and HSF2, members of the heat shock transcription factor family, activates transcription of chaperones like Hsp70 upon heat shock, proteotoxicity and oxidative stress, among other challenging conditions. The aim of this study was to elucidate the impact of oxidative stress on the protein levels of HSF1 and HSF2 and their binding to the Hsp70 promotor on DNA. K562-cells were treated with H2O2 or menadione to create oxidative stress conditions, and the relative protein levels of HSF1, HSF2 and Hsp70 were compared with Western blot. Treatment with the proteasome inhibitor MG-132 was used to examine the impact of the proteasome on the protein levels of these heat shock proteins during oxidative stress.
The results showed decreased solubility of HSF1 and HSF2 during oxidative stress, possibly due to the formation of insoluble chromatin aggregates. Oxidative stress inhibited the expected induction of Hsp70 during proteasome inhibering, both at the mRNA and protein level, by an unknown mechanism. HSF1 and HSF2 were found to be situated at the Hsp70 promotor during oxidative stress and proteasome inhibition, however, mRNA levels of Hsp70 did not increase under these conditions. Oxidative stress seems to have an inhibitory effect on the heat shock response when the cell is exposed to several combined stresses, possibly to allow transcription of other stress related genes.
